High school student Shun Takahata is bored. Bored with the day-to-day monotony of school and life, he prays for change, for something exciting. Suddenly, he and his classmates are forced to play deadly children's games and facing terrifying creatures from a talking Daruma doll to a sharp-clawed lucky cat.
Shiori Yamauchi finds a letter in her mailbox addressed to a Reiko Miura, who seems to be the previous occupant of her condo unit. She opens the letter out of curiosity. In it were the words, "I want to see you. I love you." A few days later, another letter arrived with the words, "I'll come and see you today after a long time." Scared, Shiori did an internet search for "Reiko Miura" and found out that she was the victim of a stalker murder that happened a year and a half ago.
Story continues with the students' own conflicts, Koro Sensei’s identity and the fate of the world. The time limit for assassination is approaching.
A mysterious creature destroys 70% of the moon. The creature then warns that if he is not destroyed by March of next year, Earth will be next. The creature also demands that he becomes the homeroom teacher for 3rd grade E class at Kunugigaoka Junior High School. The government is powerless to say no. The creature then becomes the homeroom teacher of 3rd grade E class which consists of failed students including Nagisa Shiota. The creature is called Teacher Koro. Meanwhile, the government requests that the students try to kill Teacher Koro even though he possesses super powers. The government offers a 10 billion yen reward for whomever successfully kills him. The students are confused by the situation, but decide to kill Teacher Koro. When their class begins with Teacher Koro, the students finds themselves having a good time with their new teacher.
In 1980, Ayuko is a new transfer student in Okayama from Tokyo. She doesn't have any friends and relies on her boyfriend Hideo who is a college student. Ayuko likes to draw a romance manga of herself and her boyfriend. Ayuko's classmate, Takemi, becomes a huge fan of Ayuko's manga and they become close friends. Due to an incident though, they drift apart. 30 years later, Ayuko is now a manga artist and comes back to Okayama to give a lecture at her former high school. There, Ayuko meets Takemi who is now a teacher.

Rino holds special feelings for Ren Kiritani, but she is unable to tell him how she feels because he is popular. One day, Ren confesses to Rino that he likes her. They begin to date and enjoy their time together, but Rino is diagnosed with ALS (amyotrophic lateral sclerosis).
Kae is a fat high school girl who is a 'fujoshi', which is girls who enjoy reading homosexual romance novels. She is dismayed when her favourite character is killed off. She is unwell and refuses to eat. She loses weight. As a side-effect she is thin and suddenly popular at school. Boys begin to pursue her. She has a choice of suitors and can pick a boy of her liking. They are all pretty.
